Among
the Critical Quality Attributes (CQAs) of interest in upstream bioprocessing
involving cell cultures is the quantitative analysis of individual amino acids
(AAs). While the one of greatest interest is typically glutamine, we have noted
interest in the quantitative analysis of various other AAs in real time in
several specific processes. Based on this interest, we have investigated the
potential to quantify multiple AAs simultaneously in solution. Being able to do
this would be very interesting for in-process monitoring and control of various
cell processes.
A Design of
Experiments was constructed and followed to determine the extent to which
individual AAs could be quantified in the presence of one another. As part of
this, some challenges were investigated with respect to the stability and
solubility of various individual AAs to determine the effect of this behavior
on the models constructed from Raman data. The employment of Net Analyte Signal
as part of this analysis allowed useful estimations of the detection limits for
each individual AA in this complex (multi-AA) matrix.
