Synopsis
High
throughput screening (HTS) is the use of automated equipment to rapidly test
thousands to millions of samples. Typically, fluorescence, bioluminescence, and
absorbance are the most commonly used assay readout technology for HTS due to
their high-sensitivity and high-speed nature. However, these techniques have a
limited chemical specificity and therefore are susceptible to false positives
and negatives. Alternatively, chromatography-based techniques are known for its
sensitivity, selectivity and quantitative analysis; however, employing them in
high-throughput screening is limited by its high run cost and slow speed. The
Polispectra® Rapid Raman Plate Reader (RPR), a newly designed/patented
Raman-based well-plate reader by HORIBA, conducts rapid chemical and/or
crystalline composition characterization on multiple samples in a 96 well plate
format for high-speed, reproducible, nondestructive, and effective screening
making it perfectly suited as an independent/reliable assay readout technology
for HTS.

we will describe how we have used RPR for high throughput quantification NTP
base; a critical step of needed for IVT reaction monitoring.
